Mostert tied himself with the Golden Lions for the 2013 season, but was seriously injured in a car crash shortly after joining, which ruled him until at least the end of 2013.

After a full recovery, he was included in the Lions squad for the 2014 Super Rugby season and made his debut the same year.

He signed for French Top 14 side Lyon ahead of the 2016–17 season on a two-year contract.

Following Pieter-Steph du Toit's injury ahead of the June Test series, Mostert was called into the Springbok squad as his replacement.

Mostert made his Springbok debut against Ireland in Johannesburg and made 5 other appearances in 2016.

Mostert's deal with Lyon fell through as the player opted to go to Japanese club Ricoh Black Rams instead, meaning that the was able to represent the Lions in the 2017 Super Rugby competition.

In March 2017 Mostert made his 50th Super Rugby appearance for the Lions against the Reds in Johannesburg.

Having already made 7 appearances for South Africa since his debut in 2016, Mostert was included in the Springbok squad for the June Test series against France.

Mostert was included in the Springbok squad ahead of the June Internationals before the 18 test cap lock signed an undisclosed contract with Gloucester Rugby.

Mostert heads into the 2018 Rugby Championship with 21 test caps.
